Why PEGylation of Peptides?
Anyone who has worked with peptide drugs has experienced that a good peptide has good activity in vitro, but as soon as it enters the body, it's gone - it's filtered too quickly by the kidneys, or it's been cleaved by proteases.
PEGylation of Peptides is the solution to this problem. By attaching a polyethylene glycol (PEG) chain to the peptide, the molecular weight becomes larger and glomerular filtration slows down; the PEG chain also blocks protease attack and reduces degradation. The result is a longer half-life and less frequent administration.
But PEG is more than just a tube to extend time. Short-chain PEGs (e.g. PEG2, PEG4, PEG8) are often used as linkers or to slightly improve solubility without changing the overall molecular weight too much. The choice of PEG depends on the effect you want.
What conditions are suitable for PEGylation?
Need linker molecule: coupled fluorescent, biotin or toxin → choose short chain PEG (PEG2/4/8)
Poor solubility: PEG is hydrophilic and improves solubility → short-chain or medium PEG is fine
Need for slow release: extended half-life of macromolecular PEG → 20k linear PEG

Three real-life cases
Case 1: A long-acting peptide program of a biotech company that
The customer had a 14 amino acid GLP-1 analog with a half-life of only 30 minutes and wanted to extend it by PEGylation, we selected a 20kDa linear PEG, attached it to the N-terminus, while retaining the activity. We chose 20kDa linear PEG, attached to the N-terminus, while retaining the activity, and the PK results showed that the half-life was extended to 12 hours after PEGylation, and one injection per day was changed to one injection per two days.
Case 2: ADC Drug Development's Connectors Program, Inc
The customer needed to synthesize a peptide containing a PEG4 linker for coupling to a toxin. Case 3: Probe optimization in a diagnostic reagent company
The customer had a peptide for the assay with poor solubility, which led to high background of the reagent. We tried three kinds of short chain modifications, PEG2, PEG4 and PEG8, and finally found that PEG4 modification increased the solubility by 3-fold, had the best signal-to-noise ratio, and did not affect the binding activity.
