Research Grade Neoantigen peptide | A key tool from target validation to mechanism discovery
Research Grade Neoantigen peptide is the most basic and central tool you can have at hand in the early stages of tumor immunology research, when you are still validating whether a newly discovered mutation can generate an effective T-cell response, or when you are screening for peptide candidates that deserve to go into follow-on development. It doesn't need to be clinical-grade endotoxin right out of the box, and it doesn't need a full set of GMP documents, but it does need to be sequenced accurately, pure, and delivered in a timely manner - because the reproducibility of your results often hinges on the quality of those few milligrams of peptide.
Science-Peptide has accumulated 20 years of experience in the field of peptide synthesis and has served a large number of tumor immunology research teams. We know what our Research Grade Neoantigen peptide customers care most: don't get stuck in the synthesis process, don't let the cell experiments go in vain because of peptide quality problems, and don't wait too long to delay the graduation of students. Therefore, we have established a set of flexible and efficient synthesis system for Research Grade Neoantigen peptide, which can be used from a few milligrams to dozens of grams, and from conventional sequences to difficult long peptides.

What is Research Grade Neoantigen peptide?
Research Grade Neoantigen peptide refers to non-clinical peptides for basic research, target validation, T-cell screening, vaccine pre-development and other non-clinical applications. Compared with clinical grade peptides, research grade peptides are more focused on synthesis success rate, delivery speed and cost-effectiveness, and are suitable for the following scenarios:
Functional validation of candidate neoantigens: Prediction software gives dozens of candidate peptides, which need to be validated one by one by ELISpot, flow or tetramer to see which ones really activate T cells.
TCR screening and affinity assay: A set of Neoantigenic peptides is needed to screen for specific TCRs or to determine the binding strength of different mutant peptides to TCRs.
Immune mechanism research: exploring how neoantigenic peptides are processed and presented, and how they activate different T-cell subpopulations.
Pre-vaccine optimization: Comparison of the effects of different lengths and modifications of peptides on the immune effect before finalizing the clinical formulation.
In these studies, Research Grade Neoantigen peptides usually come in "kits" - dozens of peptides synthesized together, a few milligrams each, with >90% or >95% purity, accompanied by HPLC and MS data are available, fast and complete.
Science-PeptideResearch Grade Neoantigen peptide core competencies
1. High-throughput synthesis, dozens of peptides delivered together
To do neoantigen research, it is often not a matter of one peptide, but dozens or even hundreds of peptides. We have a specialized high-throughput synthesis platform, which can process dozens of Research Grade Neoantigen peptides in parallel, and we can deliver all the peptides of regular length (<20 aa) to you in 10-15 working days from the receipt of the sequence to the delivery. In case of project urgency, we can compress the time by a few more days if the project is expedited.
2. Specializing in difficult peptides and not saying "can't do" so easily
The sequence of the Neoantigenic peptide is determined by mutation, not by us. We have several solutions to deal with the problem of hydrophobic, aggregation-prone, repetitive sequences:
- Microwave-assisted synthesis: For sites that are difficult to couple, microwave heating is used to improve reaction efficiency.
- Fragmentation: Long peptides of more than 30 amino acids are synthesized and reassembled by cutting them into several segments.
- Special purification system: For peptides insoluble in conventional solvents, add DMSO, guanidine hydrochloride, etc. to aid solubilization during purification.
- Suggestion for solubilization design: If the peptide is really difficult to solubilize, we can suggest to add one or two lysine at the end to improve the solubility without affecting the function.
3. Complete quality control data and traceability of experimental results
Each Research Grade Neoantigen peptide comes with a COA report containing:
- HPLC chromatogram:Purity at a glance.
- Mass Spectrometry (MS) graph: molecular weight confirmation to ensure correct sequence.
- MS/MS secondary mass spectrometry (optional): to verify the mutation site, especially suitable for experiments that need to distinguish between wild type and mutant.
This data is not only enough to write your paper, but also helps you troubleshoot experimental problems - if the results are unsatisfactory, you can at least rule out the peptide quality factor.
4. Flexible production scale and finishing options
- Scale: 1mg to 100mg is the most common, if you need a few hundred milligrams to a few grams for animal testing, we can do that too.
- Modification: we can add biotin (for tetramer), fluorescent labeling (FITC, TAMRA, Cy5, for flow-through), or carrier protein (KLH, BSA, for immunization of animals). If you need anything, just tell us in advance.
5. Intellectual property protection, your sequence only you know
The Neoantigen sequence is the core discovery of your team. All of our projects are covered by confidentiality agreements that limit project information to only a few people within the project team, and the data system is secure. We have been working together for many years and have never had a leak.

Typical application scenarios for Research Grade Neoantigen peptide
1. Functional screening of candidate neoantigens
Using predictive software to run dozens of candidate peptides from tumor sequencing data, the next step is to verify which ones really activate T cells. We have synthesized such Research Grade Neoantigen peptide kit for many laboratories, each strip is 5mg, purity >90%, directly used in ELISpot or tetramer staining. Feedback from customers is that the data is clean and there are few false positives.
2. TCR recognition profile studies
After finding a TCR that is specific for a Neoantigen, it is often interesting to see if it recognizes other similar mutations. In this case, we need to synthesize a set of mutant peptides by replacing the amino acids at key positions one by one to see whether the T cell response is still strong or not. This kind of delicate research requires high precision in peptide synthesis, which is where our MS/MS validation comes in handy.
3. Studies on the mechanisms of antigen processing and presentation
Want to study how a neoantigen peptide is cleaved by the proteasome, how it is transported by the TAP, and how it is bound by the MHC? You need to use different lengths and sequences of Research Grade Neoantigen peptide to "feed" cells or do in vitro binding experiments. We support a variety of lengths and modifications to meet the needs of mechanistic studies.
4. Comparative pre-vaccine immunogenicity
Before deciding which peptide to use in the clinic, it is often necessary to compare the immunogenicity of different peptides in mouse models. Some use long peptides, some use short peptides, and some are modified with palmitic acid. This kind of exploratory research requires quick access to peptides of different designs for comparison. We have the flexibility and speed to match.
5. Diagnostic methodology development
Some research teams are developing assays to detect neoantigen-specific T cells and need a set of Research Grade Neoantigen peptides with known positives and negatives as quality control. We can provide a small number of batches of the same peptide to ensure long-term experimental consistency.

Why Science-Peptide's Research Grade Neoantigen peptide?
1. Experience is here, and we've seen a lot of problems
We have been in this business for 20 years and have seen all kinds of Neoantigenic peptides. Some peptides, which other companies say "can't be done" when they look at the sequence, we can often find a solution after studying them for a few days. It's not that we are so good at it, it's that we have seen so many difficult peptides that we know where to start.
2. The pace can keep up with you
Students are waiting for experiments, projects are waiting for data, we can't afford the delay. We have opened a fast track for research-grade peptides, 10-15 working days for regular orders, and even faster for expedited orders. Many customers who were skeptical when they first cooperated with us have gotten used to our pace after using us once.
3. The data are complete, and the articles are sufficient
For our COA reports, HPLC and MS are standard. If you need MS/MS validation, just tell us in advance. These data not only help you to eliminate experimental interference, but also can be directly put into the Supplementary when writing the paper, so that the reviewers can not find fault.
4. Flexibility, no trouble for you
Some customers only need the crude peptide, do the purification by themselves; some customers want to split into small tubes, 1mg per tube; some customers need to ship in batches, first take a few for pre-experiment, and then make up the rest. We will cooperate with all these requirements, how to facilitate you how to do.
5. Confidentiality, your discovery is yours
The Neoantigen sequence is your core achievement. We sign a confidentiality agreement and limit the information about the project to a few people. We will not disclose any information before posting the article.
The real-life experiences of several clients that
A doctoral student of a tumor institute: he wants to validate 40 predicted neoantigenic peptides, time is tight and funds are limited. We made him 40 peptides×5mg with purity >90% and shipped them in 15 working days. He used these peptides to do ELISpot, screened 8 positives, and successfully published an article in J Immunother Cancer. later, he graduated, and his junior in the same lab asked us to do it.
Immunology lab at a university: They wanted to compare the immunogenicity of neoantigenic peptides of different lengths. We synthesized 9 peptides for the same mutation site, ranging from 9aa to 27aa, with and without biotin. The client immunized mice with these peptides and found that the 27aa-long peptide induced the strongest T-cell response, which provided key data for their subsequent vaccine design.
A couple of questions you might ask
Q:What is the difference between research grade peptides and clinical grade peptides?
A: Research grade peptides are mainly used in in vitro and animal experiments, not directly for human use. Therefore, its endotoxin and microbial limit are not mandatory, and the documents are not so thick. However, the sequence is accurate, the purity is reliable, the data can be traced, these are the same.
Q:If dozens of peptides are made together, will some of them not work?
A: We will evaluate each sequence in advance, and if we find that there is something particularly difficult to do, we will tell you in advance and suggest to change the strategy (e.g. adding solubilizing residues) or extend the cycle. For peptides of regular length, our success rate is more than 95%.
Q:What is the appropriate purity level?
A: For general T-cell assay, >90% is sufficient. For tetramer or quantitative analysis, >95% is recommended. Crude peptide (>70%) is cheaper, but impurities may stimulate non-specific T cell response, not recommended.
